Preparation of culture media and Environmental Sampling
Culture media are designed to mimic natural growth substrates, containing water, nitrogen sources (proteins, peptides), energy sources (carbohydrates), and essential growth factors like vitamins and minerals.
Solid Culture Media:
Dehydrated media were weighed, dissolved in cold and hot distilled water, and heated until transparent. The prepared media were autoclaved, with RBC plates poured after cooling.
Dilution Liquids:
Peptone and sodium chloride were dissolved in distilled water to make dilution liquids. The solution was either poured into bottles or dispensed into tubes before autoclaving.
RODAC Plate Sampling: The RODAC plate was gently pressed onto a surface for a short time, then labeled and incubated after surface disinfection.
Sponge Swab Sampling: A moistened sponge was used to swab the surface in a specific pattern to ensure thorough coverage. After sampling, the sponge was returned to its package, mixed with a diluent, and plated on PCA medium. Samples were taken before and after disinfection.
