Coomassie Staining
Interpretation
Size → protein ladder
Purity → number of bands
Amount → intensity

After running the SDS-PAGE, proteins were visualized using Coomassie brilliant blue R-250 staining, a dye that binds to proteins and produces color changes dependent on pH. Importantly, the purified sample showed only one band, indicating high purity, whereas the crude extract had two bands due to the presence of contaminants. Using the calibration equation, the molecular weights were calculated to be 13.7 kDa for the crude extract and 13.97 kDa for the purified sample. Although these values are close to the expected molecular weight, the slight increase in the purified sample’s molecular weight suggests possible modifications or interactions.
